Basically, link juices (PR percentage) are passed on when the link has no rel="nofollow" directive on it. Dofollow is the absence of rel="nofollow" in the linking code. ex. <a href="link">. So, a dofollow backlink really is a factor in increasing site's PR. Additionally, it's highly observed by SEOs that the higher the PR of the page where you get your link from, the more link juice it can pass on, the faster you'd notice an increase on your site's PR.

Right, fresh and original on page contents plus dofollow backlinks would be helpful in getting a higher PR, especially form niche relative and authority sources.

Backlinks spell the most out of PR. If you want to have high PR, you have to obtain links from web pages that have high PR themselves as much as you can (and good luck, because these high-PR backlinks are mostly hard to earn). Quality matters more than quantity, that's one golden rule in gaining PR.

Funny you asked specifically for the Dofollow links, they are the only ones important for PR (the Nofollow ones, bring you traffic only and do not improve your SERP). And, of course, qualitative content is a must - it's the characteristic of a good site.
